Tempol对UVB所致HaCaT细胞凋亡的抑制作用

摘要 目的:确定4-羟基-2,2,6,6-四甲基氧基哌啶(Tempol)对UVB所致HaCaT细胞凋亡的抑制作用。方法:HaCaT细胞分7组,A组:对照组;B组:UVB辐照组;C组:0.5 mM Tempol实验组;D组:1 mM Tempol实验组;E组:2 mM Tempol实验组;F组:4 mM Tempol实验组;G组:8 mM Tempol实验组。除A组外,其余组均辐照UVB,分析各组间HaCaT细胞增殖效应及细胞凋亡率差别。结果:与B和G组比较,各组HaCaT细胞增殖效应显著升高(P<0.01);与A组比较,(除B组外),各组HaCaT细胞凋亡率随Tempol药物浓度的升高而显著升高(P<0.01)。结论:Tempol对UVB所致HaCaT细胞凋亡有抑制作用,一定范围内其作用随Tempol浓度的升高而降低。 Objective: To determine the inhibitory effect of 4- hydroxy - 2,2,6,6- tetramethyl- piperidine 1 - oxyl (Tempol) on apoptosis caused by UVB irradiation in HaCaT cells. Methods: HaCaT cells were divided into seven groups: group A: control group, group B: UVB radiation group, group C: supplied with 0.5 mM Tempol, group D: supplied with 1 mM Tempol, group E: supplied with 2 mM, group F: supplied with 4 mM Tempol and group G: supplied with 8 mM Tempol. Except for group A, other groups were radiated with UVB, and the different cell proliferation and the apoptosis between each group were analyzed. Results: Compared with group B and G, the cell proliferation in each group was elevated significantly ( P 〈 0.01 ) ; and compared with group A, the apoptosis rate in each group was elevated significantly as the concentration of Tempol is increased except for group B ( P 〈 0.01). Conclusion: Tempol has an inhibitory effect on photodamage caused by UVB in HaCaT cells, in which the effect is decreased as the concentration of Tempol is increased in a certain range.
