摘要
目的探讨血清缺氧诱导因子-1(HIF-1)和组织P53的表达水平与乳腺癌预后的关系。方法悬浮芯片法检测117例乳腺原发浸润性导管癌、10例复发乳腺浸润性导管癌、20例乳腺良性肿瘤和31例正常人血清中HIF-1含量,并利用免疫组织化学检测其组织中P53蛋白的表达。结果原发、复发乳腺癌血清HIF-1和P53蛋白表达水平明显高于良性肿瘤和正常对照组(p<0.01),复发癌高于原发癌(p<0.05);血清HIF-1和组织P53蛋白水平在不同的病理学特征中差异有显著性(p<0.05);乳腺癌患者的HIF-1与P53表达水平呈正相关(r=0.544,=0.000)。结论血清HIF-1参与了乳腺浸润性导管癌的病理过程,是评估乳腺浸润性导管癌患者预后的重要指标。
Objective To investigate the relationship between the expression ofserum hypoxia-inducible factor 1(HIF-1α )and tissue P53 and prognosis of breast invasive ductal carcinoma.Methods Contents of serum HIF-1α in tissue and serum samples of 117 primary breast invasive ductal carcinoma,10 recurrent breast invasive ductal carcinoma,20 benign breast carcinoma and 31 healthy volunteers were detected by suspension array,and examining the expressions of P53 in them were examined by immunohistochemical method.Results The HIF-1α level and the expression of P53 in breast cancer were significantly higher than those in benign breast cancer and control group(p〈0.01);HIF-1α expression in recurrent breast cancer was significantly higher than that in primary ones in both tissue and serum samples(p〈0.05);there were significant differences of the levels of Serum HIF-1α and Tissue P53 in different pathological features(p〈0.05).Expressions of P53 were positively correlated with the content of HIF-1α(r=0.544,p=0.000) in the patients with breast cancer.Conclusion The serum HIF-1α may play an important role in the development and progression of breast invasive ductal cancer.These two markers combined can be used as an important prognosis indicator in breast invasive ductal carcinoma.
