摘要
目的对当归中药材成分的提取工艺与提取成分种类的相关性进行了研究。方法应用HPLC法测定当归提取物中的阿魏酸含量,并进行了方法学考查;以阿魏酸提取量为考查指标,探究了当归提取成分种类与超临界CO2(SFE-CO2)提取工艺条件的关系;比较了当归采用SFE-CO2提取工艺、水蒸气蒸馏工艺、水蒸气蒸馏后水提工艺、乙醇渗漉工艺、乙醇回流提取工艺等不同工艺对阿魏酸提取量的影响。结果应用HPLC法测定阿魏酸含量,精密度RSD为0.67%,稳定性RSD为1.25%,回收率达到97.35%;采用SFE-CO2提取当归中阿魏酸,以乙醇为夹带剂提取量随乙醇浓度的升高而增加,以甲醇为夹带剂提取量最大;当归中阿魏酸的提取量,以SFE-CO2提取工艺的提取量最高,依次是乙醇渗漉工艺、乙醇回流提取工艺、水提工艺,水蒸气蒸馏工艺几乎提取不出阿魏酸。结论当归药材中成分提取与SFE-CO2工艺条件密切相关;采用SFE-CO2工艺能明显提高成分的提取量且能使提取成分的种类增加,能明显提高挥发性成分的提取。
Objective To explore the relations between the extracting component variety of angelica and the extracting technology.Methods HPLC was used to determine the contents of ferulic acid in angelica extracts,and its methodology was detected.Taking the extracting quantity of ferulic acid as the subjects,the relation between the extracting component variety of angelica and the SFE-CO2 technology parameter was analyzed.The effect of different technologies,such as SFE-CO2 technology,steam distillation technology,aqua extracting after steam distillation technology,ethanol percolating technology,ethanol backstreaming extracting technology of angelica,on the extracting quantity of ferulic acid were compared.Results The RSD of precision was 0.67% by HPLC detecting ferulic acid,the RSD of stability was 1.25%,and the recovery rate was 97.35%.The extracting quantity of ferulic acid of angelica increased with the higher concentration of ethanol as an entrainer,and the extracting quantity was the maximum with methanol as the entrainer.The extract-ing quantity of ferulic acid in angelica was the most by SFE-CO2,following by the ethanol percolating technology,the ethanol recirculating extracting technology,and the aquifer extracting technology.Ferulic acid was not extracted by the steam distillation technology.Conclusion The component extraction of angelica is closely related to the SFE-CO2 technology parameters.SFE-CO2 technology can manifestly increase the extracting quantity,increase variety of extracting components,improve the extracting of volatility components,and surpass other extracting technology.
