加样时差对竞争抑制法检测抗-HBc的影响因素探讨

Study the factors of the adds the tipe time difference to the competitive inhibition method for detection of anti-HBc

目的:探讨加样时差对ELISA一步法试剂检测抗-HBc结果的影响。方法:选择抗-HBc阴性的标本分为A、B、D3组,选择抗-HBc阳性的标本设为C组。A组标本加入后放置室温不同时间再加入酶标试剂;B组标本加入后放置37℃水浴不同时间再加入酶标试剂;C组先加酶标试剂37℃水浴放置不同时间再加入标本;D组先将标本与酶标试剂混匀后加入放置室温不同时间。结果:A、B组加样方式对抗-HBc检测结果影响明显,且加标本与加酶标试剂的间隔时间越长,反应温度越高,标本的假阳性越多;C组加样方式对抗-HBc检测结果影响明显,且加酶标试剂与加标本的间隔时间越长,标本的假阴性越多;D组加样方式对抗-HBc检测结果无影响。结论:A、B、C3组实验的加样时差导致的不公平竞争可对ELISA一步法检测试剂的结果产生影响,出现抗-HBc假阳性、假阴性增多;D组加样方式可最大限度地减少抗-HBc假阳性和假阴性,保证检验质量。

Objective：Discussed the influence adds the type time difference to exame the anti-HBc result to the ELISA one-stage process reagent.Methods：Chose anti-HBc the negative sign duty A,B,D three groups,chose the anti-HBc masculine specimen as C group.After A group of specimen joins,laied aside the room temperature different time to join the enzyme sign reagent again;After B group of specimen joins,laied aside 37℃ the water bath different time to join the enzyme sign reagent again;C group added the enzyme sign reagent 37℃ water bath laying aside different time to join the specimen again first;D group first mixed uniform the specimen and the enzyme sign reagent add-on enters the laying aside room temperature different time.Results：A and B group of Canadian quadrat type resistance-HBc test result influence was obvious,and added the specimen with to add the enzyme sign reagent the time interval to be longer,the reaction tempera ture was higher,the specimen false positive were more;C group of Canadian quadrat type resistance-HBc test result influ ence was obvious,and added the enzyme sign reagent with to add the specimen the time interval to be longer,the speci men false negative were more;D group of Canadian quadrat type resistance-HBc test result did not have the influence.Conclusion：The A,B,C three groups test the Canadian type time difference causes the fair competition may have the influence to the ELISA one-stage process examination reagent result,presents the anti-HBc false positive,the false negative to increase.D group of Canadian quadrat type may maximum limit reduce the anti-HBc false positive and the false negative,the guarantee examination quality.
