摘要
目的观察穴位埋药线和穴位注射对脑缺血再灌注后大鼠纤溶酶系统PAI-1含量的影响,比较两种方法的优劣。方法将SD大鼠随机分为模型组、穴位注射组和穴位埋药线组;采用改良线栓法制备局灶型脑缺血(MCAO)再灌模型,以胶原川芎嗪制备的缓释剂(药线)在百会、大椎穴位埋药线。用免疫组化法观察各组大鼠缺血皮质区中PAI-1含量的变化。结果大鼠缺血侧皮质区PAI-1表达降低,两治疗组均能上调PAI-1的表达,穴位注射与模型组比较有显著性差异(P<0.01);其中穴位埋药线1d对PAI-1的调节作用并不明显,与模型组比较无统计学意义(P>0.05),其优势作用表现在3d和5d,与模型组比较有显著性差异(P<0.01),与穴位注射组比较有显著性差异(P<0.05)。结论穴位埋药线可以上调大鼠缺血皮质区中PAI-1的表达,减轻脑微血管基底膜和内皮细胞的损害进而保护脑微血管结构的完整性,从而起到脑保护作用,其作用优于穴位注射组。
Objective To observerve the influences of therapies of acupoint catgut-medicine embedding and injection on PAI-1 contents of plasminogen system in rats with cerebral ischemia/reperfusion injury and make comparison on superiority.Methods The SD rats were randomly divided into model group,acupoint injection group and embedding catgut and medicine in acupoints group.A modified suture occlusion method was applied to make focal cerebral ischemia(MCAO) and reperfusion model.The sustained release preparation of collagen TMP agent(drug catgut) was embedded in the Baihui(DU 20),Dazhui(DU 14) acupoints.Immunohistochemical staining was used to observe the ischemic cortex on rats PAI-1 levels change.Results The ischemic cortex was observed lowered expression of PAI-1,and the two treatment groups were observed up-regulated expression,with significant difference when compared acupoint injection group with model group(P0.01).No apparent regulative effect on PAI-1 was observed in catgut-medicine embedding for 1 day,with no statistical significance found in comparison with that of model group(P0.05).The superior effect was shown at 3d and 5d,which was significantly differed from that in model group(P0.01).Conclusion Embeded catgut can up-regulate medicine acupoint in rat ischemic cortex in the expression of PAI-1 to reduce cerebral microvascular basement membrane and endothelial cell damage and thus protect the brain microvascular structural integrity,and thus play a role in brain protection,its role is superior to acupuncture injection group
