摘要
选取540-700 nm中11个波长对不同密度的小球藻进行吸光值测定,同时对其进行精确计数和干重测定,结果表明:(1)在11个波长下,小球藻吸光值与细胞密度存在理想的线性关系,线性系数r〉0.999,其中以680 nm波长处最为灵敏,其次为540 nm;(2)小球藻吸光值与细胞干重之间同样存在良好的线性关系:Y(g/L)=0.25 X(OD680)-0.001 5,线性系数r〉0.999。表明光密度法能快速且准确地测定小球藻生物量。与传统的细胞计数法和叶绿素a测定法相比,光密度法更方便、快捷,能极大地提高实验效率和准确度。
In this paper,11 different wavelength between 540-700 nm was chosen to determine the light Absorption of different density of chlorella.At the same time,the amount and dry weight of different density of chlorella were measured accurately.The results indicated that:(1) there is a perfect linear relationship between the cell density and its optical density(OD) in the condition of 11 different wavelengths.Correlation coefficient r 0.999.The most sensitive result was under the condition of 680 nm.The second was 540 nm;(2) the relationship between OD and the dry weight of chlorella also presented a good linear relationship: Y(g/L)=0.25X(OD680)-0.001 5.Correlation coefficient r 0.999.Those results showed that optical density method can determine the Chlorella biomass rapidly and accurately.Comparing with the traditional methods,optical density method was efficiently and accurately,and providing a convenient method for laboratory research and manufacture.
