摘要
采用三氯乙酸法去除光皮木瓜粗糖中的蛋白质。利用精制后的光皮木瓜多糖测得光皮木瓜多糖对葡萄糖的换算因子后,用苯酚—硫酸法对光皮木瓜中多糖含量进行了测定,研究了显色温度、显色时间、苯酚及硫酸用量对多糖含量测定的影响。确定最佳测定条件为:显色温度100℃、显色时间15 min、苯酚用量1.0 mL、硫酸用量5.0 mL,在该条件下所测得的光皮木瓜多糖的平均含量为12.16%。且实验重现性和稳定性均较好。
In this study, crude poIysaccharides isolated from Chaenomeles sinensis was purified using 10% trichloroacetic acid solution. The content of polysaccharides was determined by phenol-sulfuric acid method and calculated from the conversion coefficient of purified polysaccharide to glucose. The optimal measure conditions were determined by investigating the relationship of UV absorbance of polysaccharides at 490 nm with action time and temperature of phenol-sulfuric acid system on polysaccharide and amount of phenol and sulfuric acid. The optimal conditions were obtained as follows: the action temperature 100℃ ,the action time 15 min,the amount of phenol 1.0 mL, the amount of sulfuric acid 5.0 mL. The average content of polysaccharides of 12.16 was obtained under the optimum testing conditions.
