摘要
目的通过对小鼠皮下肝癌进行不同温度的热生理盐水消融,探讨诱导有效免疫反应的合适消融温度。方法共120只小鼠皮下种瘤成功后,随机分为50℃、60℃、70℃、100℃及对照组共5组,每组24只,各组小鼠的皮下瘤内注射不同温度热生理盐水0.15ml进行消融,观察小鼠生存时间、肿瘤体积及消融灶内淋巴细胞和HSP70的变化。结果(1)各组小鼠生存时间不相等(P<0.05),50℃组生存时间较长。(2)各消融组体积均显著小于对照组(P<0.05);与消融前肿瘤体积比较,仅50℃缩小。(3)50℃组CD4+细胞数量显著高于其他各组(P<0.05),CD8+细胞50℃组与对照组之间差异无统计学意义(P>0.05),但高于其他热消融组(P<0.05);50℃组CD4+/CD8+比值升高最为明显;HSP70热消融组均显著高于对照组(P<0.05),50℃组表达量最高。结论50℃组消融温度可能有利于诱导产生更为强烈的主动免疫效应。
Objective To investigate the proper ablation temperature that can induce the maxium anti-tumor immunity by using different temperature saline ablation with subcutaneous liver cancer in mice. Methods After the subcutaneous tumor model was established, 120 mice were randomly divided into 5 groups, with 24 mice in each gTOUp. Except the control group, each group were given intratumor injection of 0. 15 ml saline with different temperatures respectively (50℃ ,60℃ ,70℃, 100℃ ). Mice survival time and tumor size were observed,lymphocyte number and expression of HSP'/0 in ablation foci were measured. Results (1)Survival time were different in each group(P 〈0.05 ),but the mice in the 50℃ group had the longest survival time. (2)Compared with the control group,tumor volumes in each ablation were reduced significandy(P 〈0.05),but the 50℃ group was the only group whose tumor volume reduced compared with the volume before ablation. (3)CD+ cells number in 50℃ group was significant higher than those in other groups(P 〈0.05). No significant difference of CD8+cells number was found between 50℃ group and the control group(P 〉0.05),but CD8 + cells number in 50℃ group was significant higher than those in other ablation groups (P 〈 0. 05). CD4+/CD8+ ratio increased markedly in the 50℃ group. Compared with control group,the labeling index of HSP70 in each ablation group increased significantly (P 〈0.05), and labeling index in 50℃ group was the highest. Conclusion The ablation temperature of 50℃ maybe induce a more intense active immunization antitumor effects than other temperatures.
