HLA-G和HIF-1α在上皮性卵巢癌中的表达及其临床意义

摘要 目的:探讨人类白细胞抗原-G(HLA-G)与缺氧诱导因子-1α(HIF-1α)在上皮性卵巢癌中的表达及临床意义。方法:应用免疫组化SP法检测HLA-G、HIF-1α在58例上皮性卵巢癌、30例卵巢良性肿瘤及20例正常卵巢组织中的表达,并分析它们与上皮性卵巢癌临床病理参数的关系。结果:与卵巢良性肿瘤、正常卵巢组织相比,上皮性卵巢癌组织HLA-G和HIF-1α蛋白阳性表达率都增加,差异有统计学意义(P<0.05)。HLA-G和HIF-1α阳性表达呈正相关(r=0.269,P<0.05),HLA-G阳性表达与肿瘤病理分级、临床病理分期相关(P<0.05)。结论:HLA-G和HIF-1α在卵巢癌生长、浸润、转移过程中发挥了一定作用,可作为判断卵巢组织恶性程度及评估不良预后的重要指标。 Objective:To investigate the expression of human leucocyte antigen protein G(HLA-G)and hypoxia inducible factor-lα(HIF-lα)protein in human epithelial ovarian carcinoma,and to evaluate their clinical implications.Methods:The expression of HLA-G and HIF-1α protein in 200 cases of normal ovarian tissue,30 cases of benign epithelial tumors and 58 cases of epithelial ovarian carcinoma were determined by SP immunohistochemical method,and the relationship with clinicopathological parameters were analyzed.Results:The positive rate of HLA-G and HIF-1α protein in tumor tissues was significantly higher than those in benign tumor tissues and normal tissues(60.3% and 65.5%)(P 〈0.05).There were no significant differences in HLA-G expression considering age,histology type and lymph node metastasis(P〉 0.05),while its expression was correlated with histological grade and FIGO stage Ⅲ-Ⅳ(P 〈0.05).The HIF-1α expression was significantly related with histological grade,regional lymph node metastasis and FIGO stage Ⅲ-Ⅳ(P〈 0.05),but not with age,and histology type(P 〉0.05).In additional,the expression of HLA-G was positively correlated with HIF-1α expression in epithelial ovarian carcinoma(r=0.269,P 〈0.05).Conclusion:HLA-G and HIF-1α play important roles in growth,invasion and metastasis of ovarian epithelial carcinoma,and may be a useful predictor of poor prognosis.
