摘要
选用液体发酵产纤维素酶的优良菌株里氏木霉HC-415,在摇瓶阶段,通过正交试验等对稻草培养基中稻草粉与豆粕粉的用量等因素进行了研究,确定了优化的培养基配方和产酶条件.当一级种接种体积分数为15%,一级种培养时间24 h;产酶培养基稻草粉与豆粕粉质量比为6∶1,ρ(KH2PO4)=0.01 g/mL,ρ(CaC l2)=0.006g/mL,聚醚不加,装瓶量为50 mL/250 mL瓶,培养120 h时,发酵液纤维素酶活性最高.此时,CMC ase酶活性平均为94.62 U/mL;FPA酶活性平均为16.04 U/mL.
The fine strain(Trichoderma reesei HC-415) is studied in the cellulase production by submerged fermentation.The factors in straw medium are also studied according to orthogonal experiments on shaking bottle stage.The optimized medium conditions of producing enzyme are confirmed.The cellulase activity in fermentable solution is the highest when the stair inoculation dosage is 15%,the cultured time of stair strain is 24 h,the proportion between straw and bean cake powder in medium producing celullase is 6∶1,the concentration of KH2PO4 and CaCl2 in medium respectively is 0.01 g/mL and 0.006 g/mL etc.At this time,the average CMC and FP enzyme activities are 94.62 U/mL and 16.04 U/mL.
