高效液相色谱法对鱼体肌肉组织中喹烯酮及其代谢物残留量的同时检测 被引量:5

Simultaneous Determination of Residues of Quinocetone and Its Metabolin in Muscle Tissues of Fish by High Performance Liquid Chromatography

摘要 以斑点叉尾鲴(1ctalurus punctatus)、鲤(Cyprinus carpio Linnaeus)和草鱼(Ctenopharyngodon idellus)为实验材料,建立了一种用高效液相色谱法同时检测鱼体肌肉组织中喹烯酮及其代谢物脱二氧喹烯酮残留量的方法。样品经乙腈、正己烷脱脂,旋转蒸发浓缩,乙酸乙酯洗脱,50℃氮气吹干及流动相定容后,采用Waters515高效液相色谱仪在320nm波长处检测,流速为1.0mL/min,外标法定量。喹烯酮在0.08~2.50mg/L范围内呈线性,相关系数r=0.9997,加标回收率为75%~84%,检出限为3.48~7.35μg/kg;脱二氧喹烯酮在0.02—0.50mg/L范围内呈线性,相关系数r=0.9997,加标回收率为74%~83%,检出限为5.67~16.23μg/kg。 A high performance liquid chromatographic method was developed to determine simultaneously the residues of quinoacetone and its metabolin in muscle tissues of Ictalurus punctatus, Cyprinus carpio Linnaeus and Ctenopharyngodon idellus samples. The samples were extracted with acetonitrile and defatted with hexane, concentrated by rotary evaporation, then dried under nitrogen gas at 50 ℃ and redissolved with mobile phase. The target compounds were detected by Waters 515 liquid chromatography with detection wavelength of 320 nm and flow rate of 1.0 mL/min. The quantitation analysis was performed with external standard method. The result indicated that the standard curves were linear in the range of O. 08 - 2.50 mg/L for quinocetone with a correlation coefficient of 0. 999 7, and 0. 02 - 0. 50 mg/L for desoxyquinocetone with a correlation coefficient of 0. 999 7. The recoveries of quinocetone at spiked levels of 40 - 300 μg/kg were in the range of 75% - 84% , and the detection limits were 3.48 - 7.35 μg/kg. The recoveries of desoxyquinocetone at spiked levels of 40 - 300 μg/kg were 74% -83% , and the detection limits were 5.67 - 16.23 μg/kg.
