摘要
本研究采用PCR方法从临床健康的番鸭法氏囊组织中分段扩增获得鸭圆环病毒(DuCV)PT07全长基因组,将扩增片段分别克隆,获得重组质粒,测序结果表明,DuCV基因组全长为1988nt。经基因组结构分析发现,DuCVPT07的基因组有3个开放阅读框(ORF),其中V1/rep和C1/cap是2个主要的ORF,分别编码Rep蛋白和Cap蛋白;在V1/rep和C1/cap的5'起始端之间存在与启动滚环复制有关的1个茎环结构和1个正向重复序列。遗传进化分析发现,DuCV可分为2个群,PT07与TC1/2002~TC4/2002和FJ0604同处一个进化分支,而与Ger、33753-52和MH25关系较远。
A complete genome of duck circovirus (DuCV) was amplified by PCR from bursa of a healthy muscovy duck. The DuCV (designate as PT07) genome is 1 988 nt in length containing three open reading frames (ORFs),of which the two major ORFs V1/rep and C1/cap ORF encode Rep protein and Cap protein respectively. A stem-loop structure and a directly repeat sequence element,which are indicative of a rolling-circle mode of replication,are located at the intergenic regions of the 5'-ends of V1/rep and C1/cap. The duck circoviruses could be subdivided into two clusters based on evolution analysis of DuCV gemomes. DuCV PT07 is closely related to TC1-4/2002 and FJ0604,but branched out from Ger,33753-52 and MH25 strains.
