摘要 目的:探讨仙人掌果多糖提取物(CPFP)的降血糖作用及其机制。方法:以链脲霉素诱导SD大鼠成糖尿病模型,随机分为模型组、胰岛素组、仙人掌果原汁组和仙人掌果多糖高、低剂量组并另设正常对照组。连续给药8w,观察各组大鼠体质量、血糖和血脂的变化,并用免疫组化法检测CPFP对糖尿病大鼠胰岛β细胞和α细胞的影响。结果:与模型组比较,CPFP可显著降低糖尿病大鼠血糖、胆固醇及甘油三酯,可使糖尿病大鼠体质量显著回升;免疫组化染色显示,CPFP给药组大鼠胰岛β细胞数量显著增多,但对胰岛α细胞影响不大。结论:CPFP对链脲佐菌素诱导的糖尿病大鼠有显著的降血糖和降血脂作用,其降血糖机制可能与促进胰岛β细胞分泌胰岛素有关。 Objective:To study the hypoglycemic effect and mechanism of the extracts of cactus pear fruit polysaccharide (CPFP) in diabetic rats induced by streptozotocin (STZ). Methods:The diabetic rats were induced by STZ in SD rats,and randomly divided into model group,insulin group,cactus pear juice group,high dose CPFP group,low dose CPFP group. The experimental rats were administrated for 8 weeks. During the experiment,the contents of blood glucose and blood limid of the rats were dectected and body weight were recorded. The pathology of β cell and α cell in pancreas of experimental rats were observed by immunochistochemistry. Results:Compared with modle group,the contents of blood glucose,total cholesterol and triglyceride were remarkably decreased in high and low dose CPFP groups. At the same time the body weight was significantly increased in high dose and low dose CPFP groups. The results of immunohistochemical stain demonstrated that the number of islet β cells was increased and that of islet α cells was unchanged in the treatment groups. Conclusion:CPFP can markedly decrease blood glucose and blood limid in STZ-induced diabetic rats. It′s mechanism may be related to stimulating the secretion of insulin from β cells.
