摘要
目的探讨大鼠坐骨神经移植术后BDNF在相应脊髓节段前角运动神经元内的表达变化。方法选用成年雄性Wister大鼠随机分为对照组(假手术组)和实验组(坐骨神经移植组),分别于术后不同时间点取材,应用免疫组化的方法检测相应脊髓节段前角运动神经元内BDNF的表达及其变化,根据图像分析测得的免疫强度进行统计学分析。结果BDNF在对照组脊髓节段运动神经元细胞浆内有少量表达,实验组术后可见BDNF的阳性表达的运动神经元,阳性反应为棕黄色颗粒位于细胞质。术后1周BDNF表达开始升高,2周时达高峰,6-8周后逐渐下降,至8周时仍高于对照组。结论坐骨神经移植术后BDNF在相应脊髓节段前角运动神经元中的表达增强,BDNF可能参与了神经损伤后保护神经元及促进神经再生的过程。
Objective To investigate the expression of BDNF at the motor neuron of the comu anterius medullae spinalis after the sciatic nerve transplantation in adult Wister rats. Methods Sixty adult male Wister rats were used in the experiment. Animals were randomly divided into 2 groups: control group(sham-operated group) and experimental group( nerve transplantation group). The rats were sacrificed at 3 days, 1,2,4,6 and 8 weeks postoperative and the spinal cord L4-L6 of every rat was harvested. Then we observed the altering expression of BDNF in the corresponding segments of spinal cord using imrnunohistochemieal method with image analysis system. The results were analyzed statistically. Results BDNF showed certain slight expression in control group and had no obvious changes in different phases. As to the experimental group, BDNF expression was observed remarkably in the cytoplasm of motor neuron from the 1 st week. It reached the peak in the 2nd week and gradually descended from the 6th week. There was still higher BDNF expression in the spinal cord at the 8th week compared with the control group. Conclusion The BDNF expression was activated in the motor neuron of comu anterius medullae spinalis by the sciatic nerve transplantation, which may take part in the protection of the injured motor neuron and the promafion of the nerve fiber.
