摘要
目的建立桃仁药材定性鉴别与含量测定方法。方法采用薄层色谱法以苦杏仁苷为对照对桃仁进行定性鉴别;以HPLC法对其中的苦杏仁苷进行含量测定:色谱柱:Kromasil C18;流动相:甲醇-水(20∶80);柱温:30℃;流速:1.0mL·min^-1;检测波长210nm。结果桃仁供试品的薄层色谱中,在与对照品色谱相应的位置上,显相同颜色的斑点。苦杏仁苷峰分离良好,苦杏仁苷浓度在0.033-2.116μg范围内线性关系良好,r=1.0000,平均回收率为96.11%,RSD为2.57%;结论本方法准确、可靠,可用于桃仁的定性鉴别和含量测定。
Objective To establish a qualitative identification method and a HPLC method for the determination of amygdalin in Semen Persicae.Methods TLC was employed to make qualitative identification of Semen Persicae and HPLC was used to determine the content of amygdalin in Semen Persicae: The analytical column was Kromasil C18 column,methanol-water(20∶80) as mobile phase.The flow rate was 1.0mL·min^-1 and the detection wavelength was 210nm.Results The spots in the chromatograms of Semen Persicae were in the same location and color with those in the chromatograms of reference substance.The linear range for amygdalin was 0.033~2.116μg(r=1.0000).The average recovery was 96.11%(RSD=2.57%).Conclusion The method is accurate and reliable,and it can be used in the qualitative identification and the determination of Semen Persicae.
