摘要
目的:采用高效液相色谱法对苦芪滴丸中苦参的5个单体生物碱(槐果碱、苦参碱、氧化槐果碱、槐定碱、氧化苦参碱)进行定量分析。方法:采用Inertsil NH_2(150 mm×4.6 mm,5μm)色谱柱,以乙腈(A)-无水乙醇(B)-3%磷酸水溶液(C)为流动相,梯度洗脱,流速1.0 mL·min^(-1),检测波长210 nm。结果:苦参的5个单体生物碱含量稳定。槐果碱、苦参碱、氧化槐果碱、槐定碱、氧化苦参碱进样量分别在0.14~0.49,0.38~1.30,0.06~0.20,0.05~0.18,0.13~0.44μg范围内呈良好的线性关系;r分别为0.9998,0.9997,0.9991,0.9991,0.9991。平均回收率(n=5)分别为98.2%,98.3%,97.7%,98.3%,98.9%。结论:方法可靠、实用,可用于苦芪滴丸的质量控制。
Objective:To develop an HPLC method for determination of 5 alkaloids of Radix Sophorae Flavescentis in Kuqi dropping pills.Methods:Inertsil NH2(150 mm×4.6 mm,5μm) was adopted;The mobile phase was acetonitrile (A) -absolute alcohol(B) -3%phosphonic acid water solution(C) with gradient elution at a flow rate of 1.0 mL·min^-1,and the detection wavelength was 210 nm.Results:The content of 5 alkaloids of Radix Sophorae Flavescentis was stable.The method has a good linearity in the ranges of 0.14 -0.49μg(r =0.9998) for sophocarpine, 0.38 - 1.30μg(r =0.9997) for matrine,0.06 -0.20μg(r =0.9991) for oxysophocarpine,0.05 -0.18μg(r = 0.9991) for sophoridine,and 0.13 - 0.44μg(r = 0.9991) for oxymatrine;The average recoveris were 98.2%,98.3%,97.7%,98.3%and 98.9%,respectively.Conclusion:This method is dependable,simple and practical,and may be used to control quality of Kuqi dropping pills.
