摘要
目的研究十溴联苯醚(BDE-209)在小鼠体内的富集和代谢作用。方法将健康清洁级昆明小鼠72只按体重随机分为4组,分别为对照(花生油)组、40、80和160mg/kgBDE-209染毒组,每组18只,雌雄各半。采用灌胃法进行染毒,连续染毒60d。分别于染毒结束后15、30、60d,从每组随机抽取6只小鼠,雌雄各半,测定肝脏、脾脏和肾脏多溴联苯醚(PBDEs)的含量。结果雌性小鼠在暴露160mg/kg的BDE-209达到44d以上后,其体重有增加缓慢的趋势;而雄性小鼠体重没有明显的变化。脏器对PBDEs蓄积浓度的顺序是肝脏>脾脏>肾脏,BDE-183、BDE-153、BDE-99和BDE-47四种PBDEs单体含量较高,占小鼠体内检测PBDEs总量的79%以上。结论 BDE-209进入小鼠体内会发生脱溴作用,生成低溴代联苯醚,不同脏器间PBDEs含量及组成的差别可能与其对PBDEs吸收、代谢能力的差异有关。
Objective To study the accumulation and debromination of decabromodiphenyl ether (BDE-209) in mice body. Methods Seventy-two mice were randomly divided into one control group and 3 treatment groups,18 in each, sexual ratio was 1:1, and treated with BDE-209 at the doses of 0,40,80 and 160 mg/kg, once a day, through gavage, for 60 consecutive days. The levels of PBDEs in the liver, spleen and kidney were determined at 15, 30 and 60 days respectively after exposure. Results Concentrations of PBDEs in tissues ranked as follows, liver〉spleen〉kidney. The predominant congeners found in mice were BDE-183,BDE-153, BDE-99 and BDE-47, they occupied more than 79% in the total PBDEs. Conclusion Debromination may happen in mice body exposed to decabromodiphenyl ether. Difference of concentration and components of PBDEs in different organs is associated with the ability of absorption and metabolism of the organs.
