摘要
建立了一种快速检测轮状病毒抗原的新技术——斑点酶联免疫吸附试验.用兔抗人轮状病毒IgG包被硝酸纤维素膜,然后直接浸入粪标本中反应,随后与酶标兔抗人轮状病毒IgG作用,再用底物显色,根据斑点颜色深浅,肉眼即可判断阳性或阴性结果,不需要任何特殊仪器设备.将该法与常规ELISA及聚丙烯酰胺凝胶电泳进行了比较,结果表明,与常规ELISA结果的符合率为98.11%,与聚丙烯酰胺凝胶电泳的符合率为90.57%,而且更为快速、敏感、经济、简便,易于在基层推广和现场应用.
A new technique, dot-enzyme-linked immunosorbent assay for rapid detection of rotavirus, was established. The nitrocellulose membrane (NCM) was coated with rabbit anti-human rotaviras IgG, and after it was impregnated with faecal suspensions, the NCM reacted with a peroxiodase-conjugated anti-human rotavirus IgG, and subsquently detected by the substrate. The test results were read visually according to the colour intensity without any special equipment. The new technique was found to have an overall agreement of 98.11% with conventional ELISA and of 90.57% with PAGE. It is more sensitive, rapid, simpler and cheaper than the conventionai assay and can be applied to field studies.
