摘要
目的尝试构建基于生物热力学表达的不同种质忍冬的质量评价方法。方法以金银花和灰毡毛忍冬为例,利用微量量热法,测定其作用于大肠杆菌生长代谢过程中的热功率图,建立生物热动力学模型,分析热动力学参数、生物活性与化学成分之间的相关性。结果在不同种质忍冬的作用下,大肠杆菌生长代谢的热功率图不尽一致,呈现较明显的指纹特征。不同种质忍冬作用于大肠杆菌生长代谢的热动力学参数、生物活性与化学成分之间具有显著的相关性,其中以含有机酸最高的灰毡毛忍冬的抑制作用最强。结论具有实时、在线、微量、高效、高通量、普适性好等特点的生物热力学方法和指标,既可以间接地反映中药的生物活性,也可作为评价中药质量的一种新方法。
Objective To establish a set of methods of quality evaluation of various lonicerae based on biothermodynamics. Methods Using microcalorimetry, the thermal power of Escherichia colimeta-bolism affected by Lonicera japonica Thunb and Lonicera macranthoides Hand.-Mazz.was measured. According to the thermokinetic model, the relationship between the thermokinetic parameters, caloric bioactivity and chemical constituent was analyzed.Results There was obvious difference in the caloric active fingerprints of E.colimetabolism affected by Lonicera japonica Thunb and Lonicera macranthoides Hand.-Mazz. There was obvious correlation between the thermokinetic parameters of E.colimetabolism affected by different germplasm of lonicerae, the caloric bioactivity and chemical composition. Especially, Lonicera macranthoides Hand.-Mazz with the highest content of total organtic acid showed a significant inhibitory effect on E.coli. Conclusion The thermokinetic parameters can directly reflect caloric bioactivity of different germplasm of lonicerae. As a method of biothermodynamics that is online, microamount, high-throughout, and universally-applicable,microcalorimetry is considered a useful method for quality evaluation of Chinese materia medica.
