摘要
克隆并测定福建省五条蚋Simulium(Simulium)quinquestriatum和黄毛纺蚋Simulium(Nevermannia)aureohirtumnr DNA—ITS序列,分别与GenBank中发表的Simulium亚属和Nevermannia亚属序列进行系统进化树分析,探讨其在蚋类分子分类中的作用。结果表明,各蚋种克隆株ITS2序列均与相应物种聚类,符合形态学鉴定结果,可作为蚋种鉴定和近缘种类鉴别的遗传标记之一;ITS1序列在五条蚋中同源性较低(88.3%),不适合做分类遗传标记;黄毛纺蚋泉州、漳浦两地理株间存在变异。
To explore the feasibility of ITS sequences in identifying the Simuliidae insects, the nrDNA-ITS of Simulium (Simulium) quinquestriatum and Simulium (Nevermannia) aureohirtum were cloned and sequenced. Phylogeny trees were constructed with the sequences of subgenera Simulium and Nevermannia in GenBank. The results demonstrated that sequences of S. quinquestriatum and S. aureohirtum collected in Fujian were clustered with homologous species into identical branch in the phylogenetic analysis of ITS2 gene, and according with the morphological identification. The ITS2 molecular data can be used in identifying species and distinguishing from related species in Simuliidae as a genetic marker. The ITS1 sequence is of low homology in S. quinquestriatum (88.3%) , so can't serve as a genetic marker. There existed mutations in different geographic strains of S. aureohirtum from Quanzhou and Zhangpu.
