IFN-γ对MMP-2和TIMP-2在HL-60细胞中表达的影响

摘要 目的研究白血病细胞株HL-60中基质金属蛋白酶2(MMP-2)和基质金属蛋白酶天然抑制剂-2(TIMP-2)的表达及细胞因子干扰素(IFN-γ)对MMP-2和TIMP-2转录水平的调节。方法分别在HL-60细胞生长的不同时期(1~5d)收集细胞,采用半定量RT-PCR方法,分析白血病细胞株HL-60中MMP-2和TIMP-2 mRNA的表达水平;以不同浓度的IFN-γ作用于HL-60细胞,24、48h后RT-PCR检测MMP-2和TIMP-2mRNA水平的变化。结果细胞接种后24h MMP-2的mRNA水平最高。IFN-γ各个浓度组对MMP-2 mRNA表达均有抑制作用(P〈0.05),且随着作用浓度的增高mRNA表达减少;能够增强TIMP-2mRNA表达(P〈0.05)。结论明确了白血病细胞株HL-60株在转录水平表达MMP-2和TIMP-2;IFN-γ对MMP-2 mRNA水平有明显的抑制作用,而对TIMP-1mRNA水平有增强作用。 Objective To detect the expression of MMP-2 and TIMP-2 in leukemia cell line HL-60,and explore the effect of interferon-γ(IFN-γ)on their expression.Methods Reverse transcriptase-polymerase chain reaction was used to assay MMP-2and TIM P-2 mRNA in leukemia cell line HL-60 at different time(the first day to the fifth day of the cell growth),and half.quantity values of the influence on the cells after treatment with IFN-γ in different concentration and different time(24 hours,48 hours).Results The expression of MMP-2 mRNA was significantly the highest after 24 hours of the cell culture.The expression of TIMP-2 mRNA was up-regulated by IFN-γ.IFN-γ in different concentration could down-regulate the mRNA expression of MMP-2(P0.05),and as the concentration increased,mRNA expression decreased.But the up-regulation of TIMP-2 mRNA expression could not be affected by the difference of IFN-γ.Conclusion MMP-2 and TIMP-2 mRNA are expressed in leukemia cellline HL-60.IFN-γ can inhibit expression of MMP-2 and up-grutulate expression of TIMP-2.
