摘要
采用ELISA双抗夹心法,观察了氨甲喋呤(MTX)体外对类风湿性关节炎(RA)患者外周血单个核细胞(PBMC)产生IL-1和sIL-2R的影响,结果发现,MTX(10mg/L)有抑制RA患者IL-1的分泌及LPS的促分泌作用,并对sIL-2R的自发分泌及PHA诱导分泌均有抑制作用,本文结果表明MTX对RA的治疗机制可能与其降低IL-1和SIL-2R的作用有关。
In this paper, in vitro actions of MTX on spontaneois and LPS stimulated IL - 6 and PHA stimulated sIL 2R preduction by Peripheral blood mononuclear cells(PBMC) of patients with rheumatoid arthritis (RA) werestudied. MTX(10mg/L)was added to the cultures of PBMC either in medium alone(spontancous)or in the presence 10μg of LPS or 25μg of PHA. The results showd that MTX inhibited spontaneous and LPS stimulated IL-1 production by PBMC of RA patients and spontaneous and PHA stimulated sIL - 2R production. The resultsindicate that MTX can treat RA patients by its irnmunosupression.
