摘要
目的:探讨食管鳞癌患者Th1/Th2转录因子和细胞因子的表达与临床病理因素之间的关系,为肿瘤免疫治疗提供实验依据。方法:实时荧光定量PCR(RQ-PCR)检测食管癌患者及正常组外周血单个核细胞(PBMC)中转录因子T-bet mRNA、GATA-3 mRNA表达,酶联免疫吸附法(ELISA)检测血浆中细胞因子IFN-γ、IL-2和IL-4、IL-10的表达。结果:与正常对照比较,食管鳞癌患者PBMC中Th1型转录因子T-bet和血浆中Th1型细胞因子IFN-γ、IL-2的表达水平显著降低(P<0.01);Th2型转录因子GATA-3和细胞因子IL-4、IL-10的表达水平显著升高(P<0.01),并且Th1/Th2漂移程度与肿瘤的CT分期和病灶长度相关(P<0.05),而与病变的部位、病理分型及分化程度无明显相关性(P>0.05)。结论:食管鳞癌患者体内存在Th1/Th2漂移,这可能是肿瘤细胞发生免疫逃逸的机制之一。
Objective:To investigate the relation between Th1/Th2 shift and clinicopathologic factors in peripheral blood of patients with esophageal squamous cell carcinoma(ESCC),to provide experimental evidence for anti-tumor immunotherapy.Methods: To detect the expression of transcription factors and cytokines of Th1 and Th2 type in 60 patients with ESCC.To Abstract total RNA,determine the gene expression of T-bet and GATA-3 in PBMC by real time quantitative PCR(RQ-PCR),to detect IFN-r,IL-2 and IL-4,IL-10 expression in blood plasma by Enzyme-linked immunosorbent assay(ELISA).Results: Compared with 20 healthy controls,in peripheral blood of the patients with ESCC,the expression of Th1 type transcription factor T-bet and cytokines IFN-r and IL-2 were significantly depressed(P〈0.01),while expression of Th2 type transcription factor GATA-3 and cytokines IL-4 and IL-10 were significantly enhanced(P〈0.01),and there were significant correlation between the Th1/Th2 shift and the tumor CT staging and lesion extent(P〈0.05),but there was no significant relation to the lesion location,pathological type and degree of differentiation(P〉0.05).Conclusion: Th1/Th2 shift existed in the patients with ESCC,and they maybe one of the mechanisms of tumor immune escape.
