摘要 背景与目的:有研究表明膜细胞骨架组织连接蛋白埃兹蛋白(Ezrin)能够促进肿瘤转移,可与上皮钙黏素相互作用,参与肿瘤细胞的转移过程。本研究探讨Ezrin和E-cadherin在食管鳞状细胞癌中的表达及与临床病理因素之间的关系,分析其在食管鳞癌中的诊断价值。方法:应用组织芯片技术免疫组织化学法(SP法)检测76例食管鳞状细胞癌患者组织标本及癌旁正常鳞状上皮中Ezrin和E-cadherin的表达情况,结合临床相关因素进行卡方检验及非参数检验等统计学分析。结果:76例食管癌旁正常鳞状上皮中Ezrin阳性35例(46.1%),E-cadherin阳性74例(97.4%);76例食管鳞癌组织中Ezrin阳性63例(82.9%),E-cadherin阳性34例(44.7%)。Ezrin表达与食管鳞癌的浸润深度和淋巴结转移显著相关(P<0.05);E-cadherin与肿瘤分化程度和淋巴结转移显著相关(P<0.05),随肿瘤分化程度的降低,E-cadherin的表达缺失率逐渐增加。Ezrin表达升高和E-cadherin的表达缺失呈负相关。结论:Ezrin和E-cadherin均与食管鳞癌的发生、转移有关,且两者表达呈负相关。利用组织芯片技术联合检测Ezrin、E-cadherin的表达,有助于综合判断食管癌的恶性程度和转移潜能。 Background and Objective: It has been proven that Ezrin protein may interact with E-cadherin protein and take part in metastasis of tumor cells. This study was to investigate the expressions of Ezrin and E-cadherin in esophageal squamous cell carcinoma (ESCC) and their relationship with the clinicopathologic factors,and to analyze their diagnostic values for ESCC. Method: The expression of Ezrin and E-cadherin in 72 specimen of ESCC and the para-cancer normal squamous epithelium was detected using tissue array with SP immunohistochemistry. Their correlations with the clinicopathologic factors were analyzed statistically. Results: The positive rate of Ezrin was significantly higher in ESCC than in para-cancer normal squamous epithelium (90.7% vs. 46.0%,P〈0.001); the positive rate of E-cadherin was significantly lower in ESCC than in para-cancer normal squamous epithelium (27.6% vs. 97.4%,P〈0.001). Ezrin expression was related to the invasiveness and lymph node metastasis of ESCC ( P〈0.05); E-cadherin expression was related to the differentiation and lymph node metastasis of ESCC ( P〈0.05). The high expression of Ezrin was related to the low expression of E-cadherin ( P〈0.05). Conclusion: The activation of Ezrin and the absence of E-cadherin contributes to the tumorigenesis and metastasis of ESCC.
