植物逆转座子引物开发应用中的逆转座子序列分离方法

Methods for Isolating Retrotransposon Sequences Utilized for Development of Retrotransposon Primers in Plant

摘要 文章介绍了作为植物逆转座子引物开发前提序列的逆转座子序列获得的几种方法:(1)通过同源克隆法获取逆转座子的基因序列以设计逆转座子基因序列特异引物;(2)通过筛选文库、聚合酶链式反应(polymerase chain reaction,PCR)、搜寻核酸数据库、大片段重叠群测序分离全长逆转座子序列以开发逆转座子长末端重复(long terminal repeat,LTR)引物;以及利用(3)磁珠富集、(4)抑制PCR和(5)SiteFinding PCR等方法直接获得逆转座子的LTR序列以开发逆转座子LTR引物。
