摘要
目的观察携带人脂联素(AdipoQ)基因真核表达载体是否能在人正常肝细胞株L02中表达,并探讨其对体外诱导肝细胞脂肪变的预防作用及其意义,为AdipoQ在非酒精性脂肪肝病的基因治疗中奠定基础。方法将构建含人AdipoQ重组载体pEGFPN1AdipoQ转染L02细胞(L02/pEGFPN1AdipoQ组,力=10),与pEGFP—N1空质粒转染L02细胞(L02/pEGFP—N1组,力=10)以及L02正常细胞株(L02组,力=10)为对照,分别加入浓度为20μg/ml的油酸进行培养72h,用油红O染色观察细胞内脂滴,并检测细胞内甘油三酯(TG)、游离脂肪酸(FFA)和甘油的含量。采用SAS8.2软件进行统计分析,3组之间总体差异的检验采用单因素方差分析,3组之间两两比较采用SNK—q检验。结果含人AdipoQ重组载体pEGFP—N1-AdipoQ能够在L02细胞株中表达,油红O染色显示72hL02组和L02/pEGFP-N1组分别比L02/pEGFP-N1-AdipoQ组脂肪变性更为明显。L02组、L02/pEGFP—N1组和L02/pEGFP—N1一AdipoQ组,TG含量分别为(89.23±18.45)μmol/L、(78.66±16.38)mol/L和(26.58±7.65)μmol/L,FFA含量分别为(85.39±17.26)mol/L、(75.63±12.25)μmol/L和(25.37±8.73)mol/L,甘油含量分别为(62.29±13.59)umol/L、(57.35±18.43)μmol/L和(15.37±7.53)μmol/L,F值分别为50.58、59.37、34.32,雅均〈0.01。在L02/pEGFP-N1组中TG、FFA和甘油含量均明显高于L02/pEGFP-N1-AdipoQ组,t值分别为7.81、8.50、6.75,尸值均〈0.01。在L02组中TG、FFA和甘油含量均显著高于L02/pEGFP-N1-AdipoQ组,t值分别为9.39、10.15、7.54,尸值均〈0.01。TG、FFA和甘油含量在L02/pEGFPN1组和L02组之间,差异无统计学意义。结论携带人脂联素基因真核表达载体能够在L02细胞株中表达,而且具有预防肝脂肪变性的作用。
Objective To investigate the effect of adiponectin on hepatoyte steatosis. Methods L02 cells were transfected with pEGFP-N1-AdipoQ, a plasmid encoding pEGFP-adiponectin fusion protein, or pEGFP-N1. Lipid droplets in the hepatocytes were observed by oil red staining at 72 h. The contents of TG, FFA and glycerol in hepatocytes were measured. Results Compared to cells transfected with pEGFP-N1- AdipoQ plasimd, much more lipid droplets were observed in cells transfected with pEGFP-N1 plasmid. TG, FFA and glycerol contents in L02 cells and L02/pEGFP-N1 cells were significantly higher than those in L02/ pEGFP-N1-AdipoQ cells. Conclusions Overexpression of adiponectin prevent hepatocyte steatosis.
