摘要
目的:研究猪胰蛋白酶的提取工艺,提高猪胰脏的利用价值,变废为宝,减少对环境的污染。方法:实验采用Tris-HCl缓冲液为提取剂,硫酸铵盐析和对猪胰蛋白酶的提取及纯化工艺进行了研究。通过单因素实验和正交实验,对影响猪胰蛋白酶收率和活力的诸因素进行了研究,最后确定了最佳工艺条件。结果:所得的最佳工艺条件:料液比1∶5、Tris-HCl浓度0.05mol/L、激活pH8、激活时间6h。结论:所制得的胰蛋白酶比活力为6600BAEE/mg,胰蛋白酶收率为6.5%,该工艺简单,提取效率高,产品酶活高。
Objective:To study the pig trypsin extraction craft,enhance the utilization value of pig pancreas and reduce environmental pollution. Methods:With Tris-HCl buffer solution as extracting solvents,ammonium sulfate salting-out and boric acid buffer solution crystallization were used to study how to produce and purify pig trypsin. By single-factor test and orthogonal test,the factors influenced pig trypsin activity and yield were studied and the best condition was finally confirmed. Results:The best craft condition was material liquid ratio 1∶5,Tris-HCl concentration 0.05mol/L,activation pH value 8 and activation time 6h. Conclusion:The results indicated that the activity of trypsin was 6600 BAEE/mg,the yield of trypsin was 6.5%.The technique was simple and time-saving for pig trypsin preparation with high purity,activity and yield.
