小型猪复合麻醉剂对大鼠不同脑区内源性阿片肽含量的影响 被引量:3

Effect of XFM on the endogenous opioid peptides content in different encephalic region of rats

摘要 从128只Wista大鼠中随机抽取8只为对照组,其余随机均分为5个试验组用以分别测定L-Enk、M-Enk、β-EP、dynA和OFQ的含量,每个试验组又随机均分为麻醉组、恢复Ⅰ组和恢复Ⅱ组3个亚组。采用ELISA测定各脑区内源性阿片肽的含量。结果显示,腹腔注射XFM2mL/kg后,麻醉组,大脑皮层和丘脑内L-Enk、M-Enk、β-EP和dynA含量均显著增加(P<0.05或P<0.01),海马内L-Enk、M-Enk和β-EP含量均显著增加(P<0.05或P<0.01),丘脑内OFQ的含量显著降低(P<0.01);恢复Ⅰ组和恢复Ⅱ组上述各脑区内L-Enk、M-Enk、β-EP、dynA和OFQ的含量均显著恢复(P>0.05);麻醉全过程中,小脑和脑干内5种内源性阿片肽的含量均无显著变化。结果表明,XFM对不同脑区内源性阿片肽的影响可能是其产生全麻作用的重要机理之一;XFM的中枢麻醉作用可能与增加大脑皮层和丘脑内L-Enk、M-Enk、β-EP和dynA,海马内L-Enk、M-Enk和β-EP的含量,同时降低丘脑内OFQ的含量有关。 Eight rats were randomly collected from 128 Wista rats as control group,others were randomly assigned to L-Enk,M-Enk,β-EP,dynA and OFQ groups,and each group was randomly divided into three groups(n=8):anesthesia group,recoveryⅠ group and recoveryⅡ group.The content of endogenous opioid peptides in different encephalic region of rats were quantified by an enzyme-linked immunosorbert assay.In result,after the rats were injected intraperitoneally with 2mL/kg XFM,the content of L-Enk,M-Enk,β-EP and dynA in the cerebral cortex and the thalamus in the anesthesia group were increased significantly(P〈0.05 or P〈0.01).The content of L-Enk,M-Enk,β-EP,dynA and OFQ in the above encephalic regions in the recoveryⅠ group and recoveryⅡ group were revived significantly(P〉0.05).The content of the 5 kinds of endogenous opioid peptides in the cerebellum and the brainstem did not change significantly throughout the experimental period for invasive procedures.In conclusion,effect of XFM on the content of endogenous opioid peptide in different encephalic region of rats might be an important mechanism of general anesthetic.XFM administration caused the content of L-Enk,M-Enk,β-EP and dynA to increase within the cerebral cortex and thalamus,the content of L-Enk,M-Enk and β-EP to increase within the hippocampus,and the content of OFQ to decrease within the thalamus.
