摘要
目的:研究大鼠肝缺血再灌注对肝、肾和小肠的损伤,及其丹参对上述脏器保护机制。方法:将60只Wistar大鼠分为假手术组、丹参组和实验对照组,每组各20只,实验对照组采用大鼠全肝缺血30min再灌注90min模型。测定大鼠血清谷草转氨酶(AST)、谷丙转氨酶(ALT)、乳酸脱氢酶(LDH)、肌酐(Cr)、尿素氮(BUN)水平评价肝功和肾功,采用干湿比(W/D)比较小肠水肿程度,ELISA法测定血清肿瘤坏死因子-α(TNF-α)水平,HE法比较肝、肾和小肠病理形态学差异,TUNEL法测定肾、小肠细胞凋亡指数(AI)。结果:丹参组与实验对照组相比,肝功(AST、ALT、LDH)和肾功(Cr)损害显著减轻(均P<0.05);小肠水肿程度轻,肾、小肠细胞凋亡显著减少(P<0.01);肝、肾和小肠HE染色见病理损伤改变亦轻;血清TNF-α水平明显减低。结论:丹参可减轻肝缺血再灌注对多脏器的损伤。
Objective:To examine the possibility of reducing liver,renal and small intestine injurys to the rats in liver ischemia-reperfusion by dan-shen root.Methods:Sixty male Wistar rats were randomly assigned to sham-operation,saline-treated and dan-shen root-treated groups with 20 rats in each group.Rats in the danshen root-treated group received intravenous injection of 2ml of dan-shen root dilute solution,and rats in the saline-treated group,2ml of physiological saline,30 minutes before blocking the liver blood inflow.The hepatic cord including portal vein,hepatic artery except common bile duct was blocked with a nontraumatic clamp.Thirty minutes later,the blood flow was recovered by removing the clamps.Blood samples were collected for assessing serum levels of AST,ALT,Cr and TNF-α.Livers,kidneys and small intestines underwent histopathologic examination for injury severity,and TUNEL for cell apoptosis.Results:The levels of AST,ALT,LDH,Cr in saline-treated group were significantly higher than those in dan-shen root-treated group(P0.05).The situations of histological pathology in saline-treated group were worst in the three groups by observing the slices stained with hematoxylin and eosin.The ratios of apoptotic cells in saline-treated intestine was 23.41±5.64,significantly higher than those in dan-shen root-treated and sham-operation groups(12.38±2.14)and(1.55±0.11)(all P 0.01),and the ratios of apoptotic cells in saline-treated renal was 26.05±4.58,higher than those in dan-shen root-treated and sham-operation groups(7.73±2.11 and 0.74±0.51)(all P0.01)by TUNEL examination.Conclusion:Dan-shen root has potential for reducing the liver dysfunction and multiple organ failure induced by liver I/R.
