摘要
为研究P物质激活的下丘脑大细胞神经分泌神经元及其分布,将P物质注入大鼠侧脑室,用Fos癌蛋白免疫组织化学结合催产素或加压素抗血清免疫组织化学的双标记法,检测了下丘脑各大细胞神经分泌核区被激活的催产素及加压素免疫反应性神经元。结果表明:Fos免疫强反应性的神经元主要见于第三脑室周围区及室旁核,在视上核及其他下丘脑大细胞神经分泌核区仅见到少数至极少数Fos免疫反应性神经元。Fos和催产素或加压素的免疫双染色神经元主要见于第三脑周围区,由于第三脑室周的加压素样神经元较少,因此在该区域我们见到的主要为中等至较多数量的Fos和催产素免疫双染色神经元,仅有少数的Fos和加压素免疫双染色神经元。室旁核的Fos免疫反应性神经元主要分布于室旁核的小细胞部,虽与催产素及加压素免疫反应性神经元的分布区有部分重叠,但该核区却仅见到少数Fos和催产素或加压素的免疫双染色神经元。此外,在视上核、背内侧副核也可见到少数Fos和催产素或加压素的免疫双染色神经元。本研究中将P物质注射入侧脑室后诱导的Fos阳性表达的催产素及加压素样神经元在下丘脑的分布与该区域P物质样传入纤维的分布不完全吻合,但与该区域P物质样受体的分布基本吻合。以上研究结果?
Substance P was injected into the lateral ventricle of the rat.Double immunostaining for Fos oncoprotein and oxytocin(OT)or vasopressin(VP)was used to detect the expression of c fos oncoprotein in OT like immunoreactive(OF Li) or VP like immunoreactive(VP Li)neurons in every magnocellular neurosecretory nucleus in the hypothalamus.The results showed that the intense Fos Li neurons were mainly located in the periventricular area(Pev)of the third ventricle,the preoptic area and the paraventricular nucleus(PVN)and also a few Fos Li neurons in the supraoptic nucleus (SON)and the medial dorsal accessory nucleus(MDAN).However,Fos and OT double immunostaining neurons were primarily seen in the Pev.Because relatively sparse VP Li neurons located in the Pev,in this area,only a few Fos and VP double immunopositive neurons could be identified. In the PVN,Fos Li neurons chifely distributed in the parvocellular part of this nucleus.Although there were some overlapped distribution between the Fos Li neurons and OT Li or VP Li neurons in the PVN,Fos/OT or Fos/VP double immunostaining neurons were rarely seen in this region.Moreover,sparsely distributed Fos/OT or Fos/VP double labeled neurons could be seen in the SON and MDAN.In the present study,after SP was injected into the lateral ventircle,the distribution of the induced Fos/OT or Fos/VP neurons was not entirely identity with the distribution of the SP afferent fibers in the hypothalamus,but similar to the distribution of the SP receptors in these areas.These results suggested that in the hypothalamus the regulation of SP on the OT Li or VP Li neurons might be brought about by the modulation of part of the OT Li or VP Li neurons in the Pev and a few OT Li and VP Li neurons in the PVN,SON and MDAN.
