低分子肝素对胃癌SGC-7901细胞株生长和VEGF表达的影响

Inhibitory effect of Low-Molecular-Weight-Heparins on gastric cancer cell line and the expression of VEGF

目的:探讨低分子肝素对胃癌SGC-7901细胞株的生长以及其血管内皮生长因子(VEGF)表达的影响。方法:采用MTT方法观察一定浓度速碧林(低分子肝素钙注射液)作用于SGC-7901细胞12h,24h,48h后细胞的生长情况。用逆转录聚合酶链反应(RT-PCR)分析SGC-7901细胞VEGF mRNA表达的变化,同时用酶联免疫吸附实验(ELISA)检测细胞上清中VEGF的蛋白表达量。结果:速碧林对SGC-7901细胞的生长有抑制作用,且呈剂量和时间依赖性;与对照组相比,速碧林能降低SGC-7901细胞VEGF mRNA以及蛋白的表达。结论:低分子肝素能抑制SGC-7901细胞的生长以及VEGF的分泌。

Objective：To explore the effect of Low-Molecular-Weight-Heparins on cell proliferation and the expression of vascular endothelian growth factor（VEGF） in human gastric cancer cell line. Methods ： SGC-7901 cells were cultured in 1640 medium supplied with different concentration of Low-Molecular-Weight-Heparins. The proliferative activity of SGC-7901cells were evaluated by MTT assay. The expression of VEGF in the cells were determined by RT-PCR and enzyme-linked immunosorbent assay（ELISA）. Results ：Cell proliferation were inhibitated in time dependent and dose-dependent, the expression of VEGF were reduced in the cells treated with Low-Molecular-Weight-Heparins. Conclusion ： Low-Molecular-Weight-Heparins can inhibit the proliferation of SGC-7901 cells and the expression of VEGF.
