摘要
目的探索覆盖三度烧伤切痂创面的新方法。方法酸溶解法提取鼠尾腱Ⅰ型胶原,与6-硫酸软骨素混合,制成人工真皮。该人工真皮灭菌后,移植于兔背部全层皮肤缺损创面,外用辐照猪皮覆盖保护。移植后2周,去除表面的辐照猪皮,移植自体表皮细胞,外用辐照猪皮保护。结果制成的人工真皮为半透明薄膜,内部为有孔的网状结构。移植于兔全层皮肤缺损创面后2周,人工真皮已被血管化,复合移植后2~4周,表皮细胞已分化增殖使创面愈合。结论人工真皮与自体表皮细胞复合移植方法覆盖三度烧伤切痂创面是可行的。
Objective To explore a new procedure for the covering of the full-thickness defect wound. Methods Type-Ⅰ collagen from rat tail tendon was extracted with acetic acid and mixed chondroitin-6-sulfate to form a eollagen-chondroitin-6-sulfate memhraneartificial dermis. The dermis was grafted to rabbit's full-thickness defeet wounds and overlaid with irradiated porcine skin. Two weeks later, the outer porcine skin was removed and grafts of auto-epidermal cells performed. Results The artificial dermis had a network structure with pore size about 100-150 μm and was populated with fibroblasts and vessels from wound bed following grafting two weeks. Two to 4 weeks later,the epidermal cells were differentiated and prolif- erated to heal the would. Conclusion Composite grafts of the dermis with auto-epidermal cells success-fully make the full-thickness defect wound healed.
