藏药雪灵芝的毒理学研究 被引量:11

TOXILOGICAL TEST OF TIBETAN DRUG-ARENARIA WEILI FENG WANG LIYAN, PENG BAOZHU, LI LIYING

摘要 雪灵芝的急性毒性:小白鼠半致死量(LD_(0));iv20.57g/kg、ip12.58g/kg、sc28g/kg。雪灵芝的长期毒性:选用大鼠和犬两种动物,大鼠有高、中、低三个剂量组,犬只有高低两个剂量及一个对照组。大鼠sc,犬口服给药,连续给药4周。观察指标有肝功(GPT、P_(450))、肾功(BUN)、外周血细胞计数及白细胞分类、体重增长情况以及心、肝、脾、肺、肾、脑、胸、胸腺、肾上腺等组织学镜检,并与对照组比较。雪灵芝的毒性很小,与对照组比较无显著性差异,并对肝脏有保护作用。每天每鼠口服给于雪灵芝0.5g,连续给药7个月,测定肝功(GPT)及组织学镜检,给药组与对照组比较无显著性差别。经组化检查,试验组比对照组糖元颗粒增多且个大,说明雪灵芝有保肝作用。雪灵芝的致畸实验;雪灵芝对小鼠、大鼠配对饲养,连续给药一个月,每只母鼠均怀孕,直至自然分娩,观察子代无畸形,生长发育良好。 I. Acute toxicity test of Arenaria. iv: LD50 = 20.57 ( 19.55-21.61 ) g/kg ip: ID50=12.58 ( 10.27-15.41 ) g/kg sc. LD50 = 28.0 ( 24.0-33.0 ) g/kg IILong-time toxicity test of Arenaria: Two kinds of animals rats and dogs-were chosen for test of arenaria.Three different group doses, i.e.large, moderate, and small were given to therats;only large and small were given to the rats;only large and small were given to the dogs.Administ rative route of the drug: Rat and dog is sc and per os separativeiy continued for four weeks and observe the following four indices. THey are the function of live r ( GPT.P450)and kidney ( B U N ) both R B C and w B C differential count, body weight and histologic study of heart, liver.spleen lung, kidney.brain. thymus and adrenal and compare the results with those of the control group. It was found that Arenaria has less toxicity. Yet it has protactive action of the jiver. Rats were divided into two groups. Testing dose for svery rat was O.5g of Arenaria every day continued for seven months. Measures and histologic study were also done. Experimental group was compared with the contral group. No significant difference was found. Through examination of histochemist ry, administrative drug group had more and bigger glycogen granules. while comparing with those of the control group. This indicates Arenaria has the action ofprotaction of the liver. Teratogenicity test of Arenaria: Mating of rats and mice:Each pair wrre given Arnaria for one month. The females of them were all pregnant and deiiveeed naturally. Their offsprings all had no teratogenity and developed wen.
