摘要
目的:了解骨折愈合过程中骨形成蛋白(BMP)和骨痂细胞DNA含量的变化及接骨冲剂对其的影响。方法:接骨冲剂为十二种中药的提取物。用HE、S-P免疫组化、Feulgen染色和图像分析仪测量,观察大白兔桡骨骨折愈合过程中各种骨痂成分、BMP和细胞DNA含量的动态变化,以及接骨冲剂治疗组与对照组之间的差别。结果:骨痂细胞DNA含量高峰在骨痂形成第1周;用药组细胞DNA含量高于对照组。骨痂组织BMP含量高峰在第2~4周,用药组BMP含量高峰比对照组提前1周。用药组软骨骨痂少,骨性骨痂形成及成熟比对照组提前1周。结论:DNA含量与骨痂组织增生状态有关,BMP含量与骨性骨痂形成相关。接骨冲剂能促进骨痂组织增生。
Purpose To find out the content changes of BMP and DNA in osteotylus cells and the effect of knitting granule on the changes in the course of fracture healing. Methods The granule is composed of the extracts of 12 Chinese drugs. By using HE and S P immunohistochemistry, Feulgens staining and the image analysis, observation was made on the chronological changes of various osteotylus compositions and contents of BMP and cell DNA in the course of the union of broken radii of rabbits. The differences between the group treated with the granule and the control one were also observed. 〖WTHZ〗Results〓In the first week when the DNA content in osteotylus cells reached its peak in the formation of osteotylus, the treated group had higher DNA content in the cells than the control one. For the BMP content in the osteotylus tissue which reached its peak during the second through fourth weeks, it was one week earlier for the treated group to have BMP content peak than the control one. There was less chondral osteotylus in the treated group, and the formation and maturity of osseous callus were one week earlier than those in the control group. Conclusion DNA content is relevant to the proliferation of osteotylus tissue while BMP content is relevant to the formation of osseous callus. The granule has the function of stimulating the proliferation of osteotylus tissue and the direct transformation of fibrous calluses to osseous calluses.
