摘要
为了提高橡子资源综合利用程度,对橡子壳色素提取技术进行了实验研究。通过单因素试验和正交试验L9(34)确定了橡子壳色素提取的最适条件。单因素试验结果表明,提取橡子壳色素的适宜条件是:提取剂为45%乙醇,料液比为1:40(g/mL),提取温度为80℃,时间为5h。用非极性溶剂(如石油醚)处理橡子壳粗粉可提高色素提取效果。正交试验结果表明,提取橡子壳色素的最适条件组合是:提取剂为40%乙醇,料液比为1:50(g/mL),温度为70℃,时间为5h。所得橡子壳色素是一种由多种成分组成的红褐色粉末。此外,对橡子壳色素紫外—可见分光光度法检测的工作波长进行了研究,结果表明,在390nm处,色素浓度与吸光值具有良好线性关系,回归方程为y=3.25226x+0.01288(r=0.99955),故选用390nm作为检测工作波长。
In order to improve the utilization of acorn,the acorn shell pigment extraction technology was studied.The optimum extraction conditions for the acorn shell pigment were studied by the experiment of single factor and orthogonal design.The single factor experiment results showed that the optimum extraction conditions were:the extractant was 45% ethanol;the material/solvent ratio was 1:40(g/mL);the temperature 80℃ and the time 5h.Using non-polar solvents(e.g.petroleum ether)dealing with the acorn coarse powder could increase the effect of pigment extraction.The orthogonal design L9(3^4)results showed that the optimum extraction combination of conditions was:40% ethanol as extractant,with material/solvent ratio,1:50(g/mL);temperature,70℃ and time,5h.The pigment was a red-brown powder which was consists of many kinds of ingredients.In addition,the operating wavelength of measure acorn shell pigment using UV-visible spectrophotometer was studied,the result showed that there was a good linear relationship at 390nm between the pigment concentration and the absorbance,the regression equation was y=3.25226x+0.01288(r=0.99955),therefore selected 390nm to take the measure operating wavelength.
