摘要
目的:探讨去纤维蛋白原酶预防动脉成形术后再狭窄的机理。方法:用去纤维蛋白原酶治疗刚接受血管成形术的实验兔7天,应用原位分子杂交技术,测定其腹主动脉平滑肌细胞中组织型纤溶酶原激活物mRNA的表达水平,并且与未治疗对照组比较。结果:损伤后7天对照组内弹力板附近平滑肌细胞组织型纤溶酶原激活物mRNA表达阳性,而未损伤的正常血管壁平滑肌细胞中该mRNA表达阴性;与对照组相比,去纤维蛋白原酶处理能明显减低损伤平滑肌中该mRNA表达。
Purpose To study mechanism of prevention of restenosis following angioplasty by defibrinogenase. Methods Rabbits were treated with of without defibrinogenase immediately after angioplasty for seven days. Expressions of t PA mRNA in smooth muscle cells in abdominal aorta were detected with in situ hybridization and compared. Results Expression of t PA mRNA was increased in the smooth muscle cells near tunica intima after impaired for 7 days. Treatment of defibrinogenase decreased the expression of t PA mRNA significantly. Conclusion Treatment of defibrinogenase could inhibit expression of t PA mRNA in impaired artery smooth muscle cells, thus leading to the prevention of cell proliferation and migration during restenosis.
