摘要
PEG/LiAC法转化酵母适用于酵母双杂文系统的大量筛选,并能获得106转化子/μg质粒以上的转化率,但如果没有进口的PEG3350而采用分子量接近的国产PEG4000进行转化的话,转化率仅为104,远低于文献报道的水平。我们报道了一个改进的PEG/LiAc方法,采用国卉PEG6000,转化率也能达到106,且能将热激时间从20min缩短为2min。
Metbod for yeast transformation with PEG/LiAc is very suitable for large - scale screening in two -hybrid system. tbe efficiency is more than 106 transformants/μg plasmid DNA. But the poor efficiency obtained with domestic PEG4000 instead of imported PEG 3350 is only 104 much more lower than reported by Gietz R. D. et al in 1995. Here we report a modified method which used domestic PEG6000, It can also obtained more than 106 transformants /μg ptalmid DNA transformation emciency. and shorten the heal shock tirne from 20minutes 10 2 minutes with little reduction in transformation efficiency.
