E6AP与HPV16、18蛋白表达的关系及其在宫颈癌组织中的表达及意义 被引量:2

Relationship among E6AP,HPV16,18 protein expression and their expression levels and significance in cervical carcinoma

摘要 目的:探讨E6AP基因与HPV16、18蛋白表达的关系及其在宫颈癌发生、发展过程中的作用和意义。方法:在40例宫颈癌、10例CIN及10例正常宫颈组织中采用免疫组化检测宫颈癌组织中HPV-16、18型E6蛋白表达,RT-PCR技术检测E6AP基因的表达。结果:宫颈癌组织中HPV-16、18型E6蛋白的阳性表达明显高于CIN组织和正常宫颈组织,CIN组织中的阳性表达高于正常宫颈组织,差异均有统计学意义。E6蛋白阳性组织E6APmRNA的表达(0.0946±0.0715)明显高于阴性组织中的E6APmRNA表达(0.0062±0.0103),差异有统计学意义。E6AP基因在癌组织中表达高于CIN组织及正常宫颈组织(P<0.05),E6AP的表达水平随临床分期的进展而升高(P<0.05)。结论:E6AP基因的表达与高危型人乳头瘤病毒HPV16、18E6蛋白的表达具有密切的关系,参与了宫颈上皮癌变的发生、发展过程,与宫颈癌的发生、发展密切相关。 Objective: To explore the relationship among E6AP, human papillomavirus (HPV) 16, 18 protein expression and their function and significance in occurrence and development of cervical carcinoma. Methods: The expression levels of HPV16, 18 type E6 protein in 40 cases of cervical carcinoma, 10 cases of cervical intraepithelial neoplasia (CIN) and 10 cases of normal cervical tissues were detected by immunohistochemical method, the expression of E6AP gene was detected by RT - PCR. Results: The positive expression rates of HPV16, 18 type E6 protein in cervical carcinoma were significantly higher than those in CIN and normal cervical tissues, and the positive expression rates of HPV16, 18 type E6 protein in CIN were significantly higher than those in normal cervical tissues, there was significant difference. The expression level of E6AP mRNA in E6 protein positive tissues was (0. 094 6 ± 0. 071 5 ), which was significantly higher than that in E6 protein negative tissues (0. 006 2 ±0. 010 3 ), there was significant difference. The expression level of E6AP gene in cervical carcinoma was significantly higher than those in CIN and normal cervical tissues ( P 〈 0. 05 ) , the expression level of E6AP gene increased with the advancement of clinical stage ( P 〈 0. 05 ) . Conclusion: The expression of E6AP is related to HPV16, 18 protein expression closely, which might play a role in occurrence and development of CIN, and may be related to occurrence and development of cervical carcinoma.
