摘要
采用RP-HPLC法测定发酵液中交沙霉素的含量.以C18为固定相;以甲醇-0.02 mol/L KH2PO4溶液(80∶20)为流动相,用1 mol/L的磷酸调节pH值为3,检测波长232 nm,在进样10μL、交沙霉素含量为31.25-800μg/mL范围内,交沙霉素峰面积与浓度呈良好的线性关系(r=0.9966),测试样品(n=6)RSD为0.45%,平均回收率为99.18%(n=6),RSD=0.49%.在该色谱条件下,采用RP-HPLC法能将交沙霉素与发酵液中的杂质组分实现良好分离.
This study established a method for determining the content of josamycin in fermented medium by RP-HPLC.RP-HPLC was used with C18 column as stationary phase.The mobile phase was CH3OH-0.02 mol/L KH2PO4(80∶20).The pH was adjusted to 3.0 by 1 mol/L H3PO4.The detecting wavelength was 232 nm.As a result,the calibration curves were well linear with a range of 31.25~800μg/mL(r=0.996 6),the RSD of the test samples was 0.45%,and the mean recovery rate was 99.18%(n=6),RSD=0.49%.The chromatogram system of assay is simple and reliable,and can well separate Josamycin and other impurities in fermented medium.
