摘要
目的:用分光光度法研究门冬氨酸与2,3-二氯-5,6-二氰-1,4-苯醌(DDQ)的荷移反应。方法:门冬氨酸与DDQ于pH9.5的三酸缓冲溶液中,60℃水浴中加热15 min,可生成稳定的荷移络合物。结果:荷移络合物的组成比为1:1,λmax=342 nm,表观摩尔吸光系数ε=3.42×103L/mol.cm,在10~30μg/ml的范围内符合比尔定律,回收率为97.1%~101.7%,相对标准偏差在2.9%以内。结论:应用本方法测定门冬氨酸样品的含量,结果与文献方法一致。
Objective:To develop a spectrophotometric method for determination of aspartic acid.Methods:It was based on a charge transfer reaction of the studied drug with 2,3-dichloro-5,6-dicyano-1,4-benzoquinone(DDQ) in B-R(pH9.5) buffer solution at 60℃ of water-bath for 15 min.Results:The composition of the charge transfer complex was 1:1.The apparent molor absorptivity was determined to be 3.42×103 L/mol·cm at 342 nm,and Beer′s law was obeyed in the range of 10~30 μg/ml.The relative standard deviation was less than 2.9% and the recovery was in the range of 97.1%~101.7%.Conclusion:The developed method could be applied successfully for the determination of aspartic acid in its pharmaceutical dosage forms with a good precision and accuracy compared to official methods.
