2-甲氧基雌二醇对骨髓瘤细胞作用机制的实验研究

摘要 目的 探讨2-甲氧基雌二醇(2-ME)对骨髓瘤细胞作用的机制.方法 对1例复发性和难治性骨髓瘤细胞进行分离、培养,予0、1、2、4、8和16 μmol/L的2-ME对骨髓瘤细胞处理48h(以0 μmol/L的2-ME作为对照组).后采用酶联免疫法检测培养上清中的血管内皮细胞生长因子(VEGF)和IL-6的表达水平,同时采用免疫组化法检测c-myc和bcl-2蛋白阳性表达的骨髓瘤细胞.结果 随着2-ME浓度的增加,培养上清中VEGF和IL-6的表达水平均较对照组明显降低(均P〈0.01),且骨髓瘤细胞中c-myc和bcl-2蛋白表达阳性率均较对照组明显降低(均P〈0.01).结论 2-ME对难治性骨髓瘤患者骨髓瘤细胞具有抑制VEGF、IL-6分泌和C-myc、bcl-2蛋白表达水平的作用. Objective TO investigate the effect of 2-methoxyestradiol (2-ME) on multiple myeloma cells. Methods Multiple myeloma cells were separated from a relapsed and refractory patient. The cells were incubated with 2-methoxyestradiol (0, 1, 2, 4, 8 and 16 p mol/L) for 48h, and then the contents of vascular endothelial growth factor (VEGF) and interleukin-6 (IL-6) in the supematant were detected by enzyme linked )mmunosorbent assay (ELISA). The expressions of c-myc and bcl-2 proteins in myeloma cells were determined by immunohistochemisry. Results Compared with the controls, the contents of VEGF and IL-6 in the supernatant were decreased, the expressions of c-myc and bcl-2 proteins in myeloma cells were reduced, with 2-ME concentration increased and exposure time prolonged (both P〈0.01). Conclusion 2-ME can decrease VEGF and IL-6, and down-regulate c-myc and bcl-2 protein expressions.
