HOXA10在子宫内膜样腺癌组织的表达及其临床意义

摘要 目的探讨同源框基因A10(homoboxA10,HOXA10)在子宫内膜样腺癌发生发展中的作用。方法取高分化子宫内膜样腺癌(H组)48例和中、低分化子宫内膜样腺癌(L组)30例为研究组,取15例增生期子宫内膜组织为对照组。采用原位杂交、免疫组织化学和逆转录聚合酶链反应(reverse transcription-polymerase chainreaction,RT-PCR)的方法观察HOXA10、雌激素受体(estrogen receptor,ER)、孕激素受体(progesteron receptor,PR)、p53的表达。结果①原位杂交显示H组HOXA10 mRNA呈强阳性表达,L组呈弱阳性或阴性表达,对照组呈阳性表达。②免疫组化结果显示H组HOXA10、ER、PR的表达明显高于L组(P <0.01);p53蛋白的表达结果正好相反。③RT-PCR结果显示,HOXA10 mRNA的表达强度与研究组中的组织学分级有关。④HOXA10蛋白与ER蛋白、PR蛋白之间存在明显的正相关(r=0.69,P <0.01;r=0.71,P<0.01)。HOXA10 mRNA与PRmRNA呈显著正相关(r=0.55,P <0.01)。结论子宫内膜样腺癌组织中在不同组织学分级HOXA10表达明显不同,且与ER和PR密切相关,提示HOXA10可能在子宫内膜样腺癌的发病中发挥着多方面的作用。 Objective To investigate the expression and biological significance of homoboxA10(HOXA10) in pathogenesis of endometrioid adenocarcinoma. Methods Forty-eight samples of H group (histological grade 1) and thirty samples of L group (histological grade 2 or 3) taken from patients with endometrioid adenocarcinoma were assigned into research group,and fifteen samples of proliferative endometrium as control group. Immunohistochemistry, in situ hybridization and reverse transcription-polymerase chain reaction(RT-PCR) were used to examine the expression of HOXA10, estrogen receptor(ER), progesterone receptor(PR) and p53 in the samples. Results ① Positive expression of HOXA10 mRNA was found in samples of H group by in situ hybridization, and that of L group tissue was weakly positive or negative. ②Positive expression of HOXA10,ER and PR in H group were significantly higher than that in L group( P d0.01) by immunohistochemistry. But the results of p53 expression were opposite. ③ HOXA10 mRNA expression was correlated with histological degree in research group. ④ There were positive correlations between HOXA10 protein and ER protein ( r = 0, 69, P 〈0.01), between HOKA10 protein and PR protein ( r =0.71, P 〈0.01) ,and between HOXA10 mRNA and PR mRNA ( r =0.55, P 〈0.01). Conclusion Different expression of HOXA10 in different grade of endometrioid adenocarcinoma indicates that HOXA10 may be related to the pathogenesis of endometrioid adenoeareinoma.
