摘要
目的:观察姜黄素(curcumine)对过氧化氢诱导的大鼠心肌细胞MCP-1表达的影响,探讨姜黄素在心肌细胞氧化应激中作用的可能机制。方法:以H2O2处理心肌细胞,构建心肌氧化损伤模型。并用姜黄素及H2O2共孵育心肌细胞,采用免疫印迹(Western blot)检测干预后不同时间MCP-1蛋白表达。结果:在干预后4、8、12、24、48h,H2O2组心肌细胞表达的MCP-1明显高于空白对照组(P<0.05),加入姜黄素共孵育后,各个时间点MCP-1的表达都明显下降(P<0.05)。结论:H2O2能诱导心肌细胞MCP-1蛋白表达,而姜黄素对H2O2诱导的心肌细胞MCP-1表达具有抑制作用,可能是其抗氧化应激的机制之一。
Objective:To investigate the effects of curcumine on expression of monocyte chemoattractant protein-1(MCP-1) induced by H2O2 in myocardial cell. To explore the possible mechanism of antioxygenation of curcumine in oxidative stress. Methods:In order to gain the oxidative injure cell model,cells myocardial with H2O2. Myocardial cells were incubated with curcumin and H2O2,then the expression of MCP-1 at different time points during the oxidative injure was detected by western blotting. Results:Western blotting showed that MCP-1 expressed in all groups. Higher level of MCP-1 expression was observed in H2O2 stimulated group compared with that in control group(P 〈 0.05). After treated with curcumin,the expression of MCP-l in H2O2+ curcumine group was reduced(P 〈 0.05). Conclusion:The expressions of MCP-1 induced by H2O2 can be inhibited by curcumine,and it may be one of the mechanisms of antioxygenation effect of curcumine.
