摘要
EST-SSR标记开发过程中经常会遇到一些问题,例如引物重复设计等。本试验对从大白菜EST序列中开发的30个多态性SSR标记产生的多态性条带进行测序,结果发现有一些标记的多态性并不是由于其所含有的简单重复序列重复次数的改变引起的,这类标记约占鉴定标记总数的13%。这些标记实际上属于EST-SCAR标记。在进行SSR变异对基因表达及功能影响方面的研究时,这类非SSR标记应该被排除。
Some issues could be encountered during the development of EST - SSR markers such as the repeated designing primers. The polymorphic bands of thirty polymorphic markers developed from EST sequences of Chinese cabbage were sequenced. It was found that the polymorphism of some markers, about 13 percent of total sequenced markers, did not derive from the variation of simple sequence repeats contained. These non - SSR markers belonged to EST - SCAR markers and should be eliminated in the research of the effect of SSR variation on the gene expression and function.
