摘要
在发现甘氨酸(glycine,Gly)是有可能成为内毒素(endotoxin,ET)拮抗剂的重要药物之一的基础上,从二个方面进一步观察了Gly拮抗ET的效应。(1)观察不同浓度的Gly对内毒素鲎试验凝胶形成的影响,发现随着Gly浓度的增高,拮抗效果越明显,005%~01%的Gly已显示出拮抗效应。半定量方法测出4gGly大约拮抗05μg内毒素;(2)观察Gly对ET诱导单核细胞产生EP的抑制效果。从兔血中提取单个核细胞,分别加入ET,ET+Gly,Gly,在37℃,体积分数为5%的CO2孵箱中培养。离心取上清,再分别经耳缘静脉注入60只新西兰家兔体内,结果显示:甘氨酸明显拮抗内毒素诱导单核细胞产生EP。单纯ET培养的上清,ΔT为(084±014)℃,TRI2为42±034,而内毒素与甘氨酸作用后再与单个核细胞培养的上清组,ΔT为(027±005)℃,TRI2为164±014(P<001),单纯甘氨酸培养的上清液对体温无显著影响,平均体温反应曲线也反映出同样结果。
Endotoxin(ET) is an isportant pathogen,how to inactivate it on its pathogenic effects is quite a task in the medical field.Based on the finding in our department that glycine(Gly) was an important antiendotoxin drug,the antagonistic effect of glycine to endotoxin was observed from two aspects. In the first,we observed the effect of glycine with various concentration on the TAL agglutination caused by ET,it was found that:with the increase of glycine concentration,the antiendotoxin effect of it became more evident,0 05%~0 1%Glycine had shown antagonistic effect,0 2% Gly could interrupt the TAL agglutination evidently.By using semiquatitive method,we found that 4g Gly can approximately antagonize 0 5 μg ET.In the second:Gly inhibited EP(endogenouspyrogen)production of monocytes induced by ET.First the mononuclear cells was separated from rabbits blood and adjust the cell concentration to 4×10 7 per ml,then added ET,ET+Gly(incubated at 37 ℃,5% CO 2,in before adding),Gly seperatly,then centrifuged and take the supernatants,then injected these different supernants i.v. into rabbits,the results demonstrated:Gly could antagonize obviously the EP production of monocytes induced by ET.in ET-incubating superant group, Δ T /℃ was 0 27±0 05,TRI 2 was 1 46±0 14,there was an evident difference statistically( P <0 01). Simple glycineincubating supernatant had no effect on body temperature and the P >0 05 compared with NS control.Mean body temperature response curves showed the same result.
