羟氯喹治疗口腔扁平苔藓前后的唾液双向电泳研究 被引量:6

摘要 目的:比较口腔扁平苔藓(OLP)经羟氯喹治疗后,唾液中差异表达的蛋白质,探讨羟氯喹治疗OLP的可能作用机制。方法:采用固相pH梯度(immobilized pH gradient,IPG)双向凝胶电泳(two-dimensional gel electrophoresis,2-DE)分离OLP经羟氯喹治疗三个月后唾液中的差异蛋白质,银染显色,Imagemaster图像分析软件分析差异蛋白质点。结果:①羟氯喹治疗OLP前和治疗后的唾液2-DE图谱的平均蛋白质点数分别为1894±183个和1920±341个,凝胶图谱分辨率、重复性较好;②通过Imagemaster图像分析比较治疗前后的唾液2-DE图谱,差异表达的蛋白质点数为21个,其中6个点在治疗3个月后高表达,15个点在治疗后低表达。结论:本研究建立了分辨率高且重复性较好的OLP治疗前后的唾液双向凝胶电泳图谱,发现存在差异表达的蛋白质,为进一步研究羟氯喹治疗OLP的作用机制奠定了基础。 Objective:To compare the difference of the serum protein about the patient of oral lichen planus who received hydroxychloroquine therapy and to explore its possible mechanism.Method:The total serum proteins of oral lichen planus who received hydroxychloroquine therapy were separated by means of immobilized pH gradient-based two-dimensional gel electrophoresis(2-DE).After AgNO3 staining,the differential expression proteins were ana1yzed by using ImageMaster image ana1ysis software.Result:(1)The well-resolved,reproducible 2-DE patterns of the serum of oral lichen planus who received hydroxychloroquine therapy were obtained.The results showed that average spots before and after the medicine treatment were 1894±183 and 1920±341 respectively;(2)The 21 differential protein spots were identified by ImageMaster image analysis software.There were 6 protein spots were higher expression、15 protein spots were lower expression after the hydroxychloroquine treatment.Conclusion:In this study,the well-resolved reproducible 2-DE patterns of the serum of oral lichen planus before and after the treatment of hydroxychloroquine were established and some differential proteins were characterized,which will provide a basis for elucidating the mechanism of hydroxychloroquine action on oral lichen planus.
