丹参多酚酸盐对氧化低密度脂蛋白促血管平滑肌细胞分泌作用的影响 被引量:9

摘要 目的:研究丹参多酚酸盐对氧化低密度脂蛋白(oxLDL)致兔血管平滑肌细胞(VSMC)分泌细胞因子的影响。方法:兔VSMC分5组处理,即对照组、oxLDL(50 mg.L-1)组和不同浓度丹参多酚酸盐(10、50、100 mg.L-1)加oxLDL(50 mg.L-1)组。以酶联免疫吸附法检测培养上清液中细胞因子白细胞介素18(IL-18)、白细胞介素1(IL-1)以及可溶性细胞间黏附分子(ICAM-1)水平。结果:oxLDL(50 mg.L-1)可促进VSMC分泌IL-18、IL-1和ICAM-1,于作用后8 h分别增至对照组的1.7、3.2和2.1倍,与对照组比较均有非常显著差异(均P〈0.01)。丹参多酚酸盐可浓度依赖地抑制oxLDL的促IL-1和ICAM-1分泌作用(P〈0.05),在50~100 mg.L-1浓度范围内对IL-18的产生具有抑制作用(P〈0.05)。结论:oxLDL可促进兔VSMC分泌细胞因子IL-18、IL-1及ICAM-1。丹参多酚酸盐可能通过抑制oxLDL刺激的细胞因子分泌对VSMC免疫学功能发挥调控效用。 Objective: To investigate the effects of oxidized low density lipoprotein(oxLDL) on cytokins secretion of rabbit vascular smooth muscle cells(VSMCs) and the intervention effect of salvianolate.Methods: Rabbit VSMCs were cultured and divided in to five groups: the contorl group,oxLDL(50 mg·L-1) group,different concentrations of salvianolate(10,50,100 mg·L-1) plus oxLDL(50 mg·L-1) group.The levels of interleukin-18(IL-18),interleukin-1(IL-1) and intercellular adhesion molecular(ICAM-1) were evaluated by ELISA method.Results: oxLDL increased the secretion of IL-18,IL-1 and ICAM-1 by 1.7,3.2 and 2.1 fold respectively compared with th control group(P0.01).Salvianolate decreased the secretion of IL-1 and ICAM-1 significantly in a concentration dependent modality(P0.05).The inhibiting effect on the secretion of IL-18 can be found when treated with 50 mg·L-1 and 100 mg·L-1 of salvianolate(P0.05).Conclusion: oxLDL can increase the secretion of IL-18,IL-1 and ICAM-1 in rabbit VSMCs.Salvianolate may regulate the immunological function of VSMCs by inhibiting the secretion of above-mentioned cytokins stimulated by oxLDL.
