摘要
番茄(Lycopersiconesculentum)愈伤组织和胡萝卜(Daucuscarota)愈伤组织经混合共培养后生长旺盛。仔细挑选嵌合组织转移至含1.0mg/LBA和0.5mg/LNAA的MS培养基上继代培养。将嵌合组织转移至含1.0mg/L2,4-D和0.5mg/LBA的MS培养基上培养3、8、28d时进行电子显微镜观察。结果表明:异质科间细胞相互作用在早期表现为隔离层形成和逐渐变薄解体,随后形成次生胞间连丝和胞间通道,在晚期还观察到相互接触的两种异质细胞的质膜、核膜和线粒体膜的消失以及异质细胞壁变薄解体。
Well mixed tomato(Lycopersicon esculentum) calli and carrot(Daucus carota) calli grew flourishingly when subjected to culture. Chimeral calli were selected carefully and transferred to Murashige and Skoog (MS) medium containing 1.0 mg/L 6-Benzylaminopurine (BA) and 0.5 mg/L Naphthalene acetic acid (NAA). Electron microscopic observations of chimeral interfamilial cells were performed after 3, 8 and 28 days of subculture of chimeral calli in MS medium containing 1.0mg/L 2, 4 - Dichol- orophenoxyacetic acid (2, 4 - D) and 0.5 mg/L BA. Formation and disintegration of isolation layers were seen at the early stage, followed by subsequent formation of secon- dary plasmodesmata and intercellular passages. Disappearance of plasma membrane, nuclear membrane and mitochondrial membrane in the heteroplasmic cells in contact then took place. Ultimately, disintegration of heteroplasmic cell walls was seen.
