摘要
目的比较结核杆菌在六种培养基上的生长速度,建立实验教学中快速培养结核杆菌的方法。方法制备六种结核杆菌培养基各20支,每支接种TB菌液(10-2mg/ml)0.5ml,观察结核杆菌生长情况。结果结核杆菌在Middlebrook 7H10培养基上生长最快,接种6天有3株出现了菜花状菌落,第13天20支培养基斜面上有19支均长满菜花样菌落,阳性率达95%,其次是新鲜马铃薯鸡蛋培养基和马铃薯粉、鸡蛋培养基在第6天出现1株和第8天出现2株典型菜花状菌落,在第27天和25天20支培养基中各有18和17支菜花样菌落均长满斜面培养基,阳性率达90%和85%。结论结核杆菌在Middlebrook 7H10培养基上生长最快,培养时间最短,其次是新鲜马铃薯鸡蛋培养基和马铃薯粉鸡蛋培养基,可作为理想的实验教学用结核杆菌培养方法。
Objective To compare the growth rates of Mycobacterium tuberculosis in 6 kinds of self-prepared culture media and to establish methods of rapid culture of Mycobacterium tuberculosis for experimental teaching.Methods Six kinds of solid media were prepared by modifying Lowenstein-Jensen culture medium and each kind had 20 tubes.Each tube was inoculated with 0.5 ml BCG suspension(10~2 mg/ml)and the growth of Mycobacterium tuberculosis was observed.Results Mycobacterium tuberculosis was found fastest growth in Middlebrook 7H10 culture media:typical cauliflower-like colony of 3 strains of Mycobacterium tuberculosis appeared on day 6 and 19 strains,on day 13,and the positive rate was 95%.Typical cauliflower-like colony of 1 strain and 2 strains of Mycobacterium tuberculosis appeared in fresh potato-egg and potato powder-egg culture media on days 6 and 8,respectively.On days 27 and 25,typical cauliflower-like colony was observed in 18 and 17 tubes of these two culture media,respectively and the positive rates were 90% and 85%,respectively.Conclusion Mycobacterium tuberculosis grew fastest in Middlebrook 7H10 culture media,followed by fresh potato-egg culture and potato powder-egg culture.All these three kinds of culture media of Mycobacterium tuberculosis may be good choices for experiment teaching.
