摘要
目的探讨姜黄素对急性出血坏死性胰腺炎环氧化酶-2(COX-2)表达的影响。方法将SD大鼠随机分成3组:假手术(SO)组、急性出血坏死性胰腺炎模型组(AHNP)、姜黄素预处理(Cur)组。Cur组在造模前2h经腹腔注射姜黄素溶液(200mg/kg),SO组和AHNP组注射等剂量的二甲基亚砜(DMSO)溶液。AHNP组和Cur组用5%(50g/L)牛磺黄胆酸钠0.1mL/100g胰胆管逆行注射建立急性出血坏死性胰腺炎模型,SO组大鼠开腹后翻动胰腺3次即关腹。3组大鼠分别于术后3、6、12h处死大鼠,每时间点各6只大鼠,观察胰腺组织病理学改变、胰腺组织COX-2表达和血清淀粉酶的变化。结果AHNP组胰腺组织COX-2表达和胰腺损伤评分进行性增加,Cur组在各时相点胰腺损伤的程度均显著减轻,COX-2的表达水平下降(P<0.05)。结论COX-2参与了急性胰腺炎的发生和发展,姜黄素可能通过抑制COX-2表达,发挥对急性出血坏死性胰腺炎的保护作用。
Objective To investigate the influence of curcumin on cyclooxygenase-2(COX-2) expression in severe acute pancreatitis.Methods SD Rats were randomly divided into 3 groups:sham-operation(SO) group,acute hemorrhagic necrotic pancreatitis(AHNP) group and curcumin-treated(Cur) group.Treated group was injected curcumin (200 mg/kg) through abdominal cavity before operation while control and AHNP groups were injected nomal-dimethyl sulfoxide.Treated and AHNP groups were operated to induce AHNP model which was produced by injecting 5%(50 g/L) sodium taurocholate at a dosage of 0.1 mL/100 g via the biliopancreatic duct,while sham-operation(SO)groups were only turned over pancreas three times after abdominal cavity opening.Rats were sacrificed at 3、6、12 hours after induced of pancreatitis(n=6).The pancreatic tissue specimens were collected to assess the pathological score.the expressions of COX-2 protein in pancreas were assayed by immunohistochemistry.Serum was collected for measurement of amylase.Results In the AHNP group,the score of histopathology and COX-2 expression in pancreas were increased significantly and time-related.In the treated group,the extent of pancreas injury and the level of COX-2 protein expression was significantly lower than AHNP group at the same time (P0.05).Conclusion COX-2 plays an important role in severe acute pancreatitis,and curcumin can down-regulate the expression of COX-2 protein,so that it can improve inflammatory injury of pancreatitis.
